Nicotinamide riboside is a vitamin B3 form that raises NAD+ in the blood. A small crossover trial in healthy middle-aged and older adults found it well tolerated.

Who was studied: Healthy middle-aged and older adults, each of whom took nicotinamide riboside and placebo in turn.
How: Randomized, double-blind, placebo-controlled crossover trial with two six-week periods.
What it found: NR was well tolerated and raised NAD+ metabolism. Hints of lower blood pressure and arterial stiffness were reported as reasons for future trials, not as proven effects.
Limits: Raising a blood marker is not the same as improving health. The trial was small and short, and the physical-function findings were exploratory.

Short-term use was well tolerated in this trial. Long-term effects remain uncertain, and people with medical conditions should ask a clinician before using NAD+ precursors.
